Dopamine receptor D2 (DRD2) is a G protein-coupled receptor (GPCR) that primarily couples to the Gi subtype of G proteins, leading to inhibition of adenylyl cyclase activity. While DRD2 is well-known for its roles in the central nervous system, it also has significant functions in the gastrointestinal (GI) tract, including motility regulation, secretion/absorption modulation, and mucosal protection. Recent research highlights its role in colonization resistance via sensing microbial tryptophan metabolites. It is expressed in both mucosal tissues and muscular layers of the GI tract.
