Overview

The dopamine receptor D2 (D2R) is a G protein-coupled receptor (GPCR) that mediates dopaminergic neurotransmission, particularly within the central nervous system. The postsynaptic D2 receptor couples to Gi/o proteins, inhibiting adenylyl cyclase activity and decreasing cyclic AMP levels. It plays a key role in regulating neurotransmitter release, synaptic plasticity, motor control, reward processing, and is a major target for antipsychotic drugs. Alterations in postsynaptic D2R signaling are implicated in schizophrenia, Parkinson’s disease, drug addiction, and ADHD.
