Overview

Dynein light chain Tctex-type 1 (TCTEX-1), encoded by the DYNLT1 gene, is a critical component of the cytoplasmic dynein-1 motor complex (UniProt: P63172). It serves as a light chain subunit that functions primarily as a cargo-binding adapter, linking specific cellular components to the dynein motor for retrograde transport along microtubules (NCBI Gene: 6993). Beyond its structural role in the motor complex, TCTEX-1 participates in various signaling pathways, including the regulation of G-protein signaling and the modulation of actin cytoskeleton dynamics (PubMed: 11500494). It is essential for processes such as neurite outgrowth, ciliogenesis, and the maintenance of Golgi apparatus integrity (PubMed: 21670209). In the context of disease, TCTEX-1 is frequently exploited by various viruses, such as Rabies and HIV-1, which bind to the protein to facilitate their transport toward the host cell nucleus (PubMed: 15548690). Additionally, TCTEX-1 is often overexpressed in several types of cancer, where it contributes to increased cell proliferation, migration, and invasion (PubMed: 25663484). While no FDA-approved drugs currently target TCTEX-1, it is an active area of research for developing small-molecule inhibitors and competitive peptides to block viral entry and tumor progression.
