Overview

E74-like factor 2 (ELF2) is a member of the ETS transcription factor family, characterized by a highly conserved helix-turn-helix ETS domain that enables sequence-specific DNA binding and the regulation of gene transcription. ELF2 modulates the activity of RNA polymerase II and functions in both the activation and repression of gene expression, depending on the isoform present (ELF2A as activator and ELF2B as repressor)[1][4][5]. It plays essential roles in cell cycle progression, apoptosis, and the differentiation of early B and T cells, as well as angiogenesis[1][2]. ELF2 has been implicated in several cancers, including hepatoma, lung cancer, and leukemia, with alterations in its expression linked to tumorigenesis and poor prognosis[1][3][4][5]. Alternative splicing results in different isoforms, which can have distinct functional effects in normal and diseased tissues[5]. ELF2 is located in the cytosol and nuclear body and interacts with other transcriptional co-regulators such as RUNX1[4]. The gene is conserved among species and has multiple aliases reflecting its historical discovery and molecular characteristics. While there is ongoing research into its use as a biomarker (especially in breast cancer), no approved drugs are known to directly target ELF2 at present[3][4].
