The glycan cap is a heavily glycosylated region of the ebolavirus envelope glycoprotein (GP) GP1 subunit. It sits between the receptor-binding domain (RBD) and the mucin-like domain (MLD), forming part of a 'chalice' structure. Its primary function is to shield critical functional domains from recognition by host antibodies, contributing to immune evasion. It is also involved in viral entry and is a major target for neutralizing antibodies derived from survivors.
Antibodies targeting the glycan cap can neutralize the virus by blocking conformational changes required for fusion or promoting clearance via Fc-mediated mechanisms.
