Overview

The target consists of eight predefined tumor-associated antigen (TAA) epitopes specifically selected for their frequent presentation on glioblastoma cells and their demonstrated immunogenicity in previous clinical trials such as IMA950 and GAPVAC. These epitopes are encoded by the CVGBM mRNA vaccine and are presented on the cell surface via HLA-A*02:01 (five epitopes) and various HLA-DR molecules (three epitopes). By presenting these specific peptides, the vaccine aims to stimulate a coordinated immune response involving both CD8+ cytotoxic T lymphocytes and CD4+ helper T cells against the tumor. This multi-epitope approach is designed to overcome the low mutational burden and high heterogeneity characteristic of glioblastoma, potentially limiting tumor immune escape. Clinical trials have shown that targeting these epitopes can induce de novo T-cell responses in a majority of patients with newly diagnosed glioblastoma, particularly those with MGMT-unmethylated status.
