Electrogenic sodium bicarbonate cotransporter 4 (NBCe2), encoded by the SLC4A5 gene, is a transmembrane protein that facilitates the movement of sodium and bicarbonate ions across the plasma membrane (UniProt Q9BY07). It is primarily expressed in the kidney, liver, and choroid plexus, where it plays a vital role in maintaining acid-base balance and regulating intracellular pH (PubMed: 22539647). In the renal system, NBCe2 contributes to sodium reabsorption, and its dysfunction or genetic variation is strongly linked to salt-sensitive hypertension and blood pressure regulation (PubMed: 15605130). Beyond the kidney, NBCe2 is involved in the production of aqueous humor in the eye, positioning it as a potential therapeutic target for managing intraocular pressure in glaucoma (PubMed: 25673775). Although no specific clinical drugs are currently approved for NBCe2, experimental inhibitors like S0859 are frequently utilized in research to modulate its activity. Targeting this transporter offers a strategy for treating cardiovascular and ocular diseases by influencing ion and fluid homeostasis.
