Overview

Endometrial tissue refers to the mucosal lining of the uterus, which undergoes cyclical changes under hormonal regulation. Suppression of endometrial tissue is a therapeutic target in conditions such as endometriosis, where endometrium-like tissue grows outside the uterine cavity and causes pain, inflammation, and infertility. Key molecular targets for suppression include estrogen receptors, progesterone receptor, matrix metalloproteinases, and immune modulation.

Mechanism of action

Suppression strategies focus on: - Blocking estrogen action via selective estrogen receptor modulators or aromatase inhibitors. - Enhancing progesterone response or mimicking its effects with progestins; however, efficacy may be limited by intrinsic progesterone resistance due to reduced PGR levels or function. - Inhibiting matrix metalloproteinases to prevent invasion/spread of ectopic tissue. - Modulating immune responses to enhance clearance of aberrant tissues.
