Overview

Endosomal Toll-like receptors (TLRs) are a specialized group of pattern recognition receptors, including TLR3, TLR7, TLR8, and TLR9, located within the endosomal compartments of immune cells such as dendritic cells and macrophages [1]. These receptors are primarily responsible for sensing nucleic acids derived from pathogens or damaged host cells: TLR3 recognizes double-stranded RNA, TLR7 and TLR8 recognize single-stranded RNA, and TLR9 recognizes unmethylated CpG DNA motifs [3]. Upon ligand binding, they initiate signaling through the MyD88 or TRIF adapter proteins, resulting in the robust production of type I interferons and pro-inflammatory cytokines that bridge innate and adaptive immunity [2]. In drug development, endosomal TLRs are targeted via nucleic acid delivery systems, such as synthetic oligonucleotides or lipid nanoparticles, to act as vaccine adjuvants or potent anti-tumor agents [4]. Conversely, dysregulated sensing of self-nucleic acids by these receptors is linked to the pathogenesis of autoimmune conditions like systemic lupus erythematosus and psoriasis, making them important targets for inhibitory therapies [2]. Consequently, both agonists and antagonists of these receptors are being explored for their therapeutic potential in oncology, infectious diseases, and autoimmunity [2, 4].

Mechanism of action

Agonism of endosomal Toll-like receptors triggers signaling through adapter proteins MyD88 (for TLR7, TLR8, and TLR9) or TRIF (for TLR3), leading to the activation of interferon regulatory factors (IRFs) and NF-κB, which results in the production of type I interferons and pro-inflammatory cytokines [1, 3].
