Enterotoxigenic Escherichia coli (ETEC) lipopolysaccharide (LPS) is a complex glycolipid and a major structural component of the outer membrane of ETEC, a leading cause of traveler's diarrhea and childhood mortality in low-income countries (Fleckenstein et al., 2010). The molecule consists of three regions: Lipid A, a core oligosaccharide, and the O-antigen; Lipid A serves as the primary endotoxic moiety that triggers the host's innate immune system via the TLR4/MD-2 receptor complex (Park & Lee, 2013). This interaction leads to the production of pro-inflammatory cytokines, which can contribute to the pathogenesis of infection. The O-antigen component is highly immunogenic and is a primary target for vaccine development, such as the ETVAX candidate, which aims to induce mucosal IgA responses (Lundgren et al., 2014). Therapeutic interventions targeting LPS include polymyxin antibiotics, which bind to the negatively charged Lipid A to disrupt the bacterial membrane and neutralize endotoxin activity (Nation et al., 2014). However, the high structural variability of the O-antigen across different ETEC strains remains a significant challenge for broad-spectrum vaccine efficacy.
