The HIV-1 Envelope glycoprotein 140 (gp140) is a recombinant, soluble form of the HIV-1 envelope spike protein, engineered to mimic the native structure of the viral surface trimer. It is a mosaic, multi-clade construct designed to represent diverse strains of HIV-1 and is widely used as an immunogen in vaccine research due to its ability to elicit neutralizing antibody responses. gp140 consists of gp120 (the surface subunit responsible for binding to host cell receptors CD4 and coreceptors CCR5 or CXCR4) and gp41 (the transmembrane subunit that mediates fusion between viral and cellular membranes). Native Env exists as a trimer of heterodimers (three copies each of gp120/gp41). Recombinant gp140 can form stable trimers that closely resemble this native conformation. As the only HIV protein exposed on virions, Env—and specifically constructs like mosaic/multi-clade gp140—is a primary target for neutralizing antibodies and vaccine design efforts.
