Overview

Envoplakin (EVPL) is a member of the plakin family of structural proteins that is integral to the formation and maintenance of the cornified envelope in keratinocytes, a key component of the skin barrier. It is expressed in the epidermal cornified envelope and may act as a link between desmosomes (cell-cell junctions) and intermediate filaments, contributing to the structural integrity of stratified squamous epithelia. The EVPL gene is located on chromosome 17q25. Deletion of this gene has been linked to both familial and sporadic forms of oesophageal squamous cell carcinoma. Additionally, patients with paraneoplastic pemphigus, an autoimmune blistering disorder, produce antibodies against envoplakin, implicating it in autoimmune pathology[1][5]. Its main paralog is periplakin (PPL), with which it can interact[1][2].
