Overview

Epidermal growth factor receptor pathway substrate 8 (EPS8) is a scaffold protein that plays a critical role in actin dynamics and cell signaling, particularly within the EGFR pathway (Offenhäuser et al., 2004). While its expression is tightly regulated in normal tissues, it is significantly upregulated in various malignancies, including colorectal, pancreatic, and head and neck cancers, where it promotes cell proliferation and metastasis (Maa et al., 2001; Waseem et al., 2007). MHC class I-presented EPS8 peptides are specific fragments of this protein that are processed and displayed on the cell surface by Major Histocompatibility Complex molecules. These complexes act as specific targets for the immune system, allowing CD8+ cytotoxic T lymphocytes to identify and eliminate cancerous cells (Tsang et al., 2004). In therapeutic contexts, these peptides are being investigated as targets for cancer vaccines and T-cell receptor (TCR) engineered therapies designed to elicit a robust anti-tumor immune response. The specificity of these peptides to tumor cells makes them attractive candidates for precision immunotherapy, although challenges such as MHC downregulation by tumors remain a concern.
