Epithelial membrane antigen (EMA), also recognized as Mucin-1 (MUC1) and CD227, is a heavily glycosylated transmembrane mucin glycoprotein found on the apical surface of glandular and ductal epithelial cells as well as some hematopoietic cells. It is used widely in immunohistochemistry to characterize epithelial origin tumors, especially adenocarcinomas, and serves as a diagnostic and prognostic marker in cancer. EMA/MUC1 plays roles in cell adhesion, signaling, tumor progression, and immune response. The molecule is associated with poor prognosis in many cancers and is targeted for experimental antibody-based therapies. Its heterogeneous and broad distribution across different tissue types introduces challenges in therapeutic targeting and diagnostic specificity[1][5][7][9][11].
