Overview

Estrogen receptor alpha (ESR1) mRNA is the transcript of the ESR1 gene (NCBI Gene ID: 2099), which encodes the estrogen receptor alpha (ERα) protein, a member of the nuclear receptor superfamily (UniProt P03372). This mRNA is essential for the translation of ERα, which functions as a ligand-dependent transcription factor regulating genes involved in cell proliferation, reproductive development, and bone homeostasis (PubMed: 29129569). In oncology, particularly in hormone receptor-positive breast cancer, ESR1 mRNA is a key driver of disease progression and a primary site for mutations that confer resistance to standard endocrine therapies (PubMed: 30333295). While most current treatments target the ERα protein, the mRNA itself is an emerging therapeutic target for RNA-based modalities such as antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) (PubMed: 31515463). These experimental strategies aim to silence the expression of both wild-type and mutant ESR1, providing a potential pathway to overcome resistance mechanisms that bypass protein-level inhibition.
