ETS proto-oncogene 1, transcription factor (ETS1) is a member of the ETS family of transcription factors characterized by a conserved ETS DNA-binding domain that recognizes GGAA/T motifs in gene promoters[1][2]. ETS1 regulates gene expression of genes involved in cell proliferation, differentiation, migration, apoptosis, immune responses, and angiogenesis[1][2]. It plays a key role in lymphoid lineage differentiation and in responses to extracellular signals via MAPK/ERK and PI3K/Akt pathways[1][2]. Aberrant expression or activity of ETS1 is linked to tumor progression, metastasis, and immune escape, making it a biomarker and potential therapeutic target in oncology[2]. ETS1 also interacts with DNA repair pathways and proteins (such as PARP1 and DNA-PK), impacting DNA damage resistance and cell survival[1]. Because ETS1 influences many vital processes in normal and diseased tissues, strategies modulating its function must account for complex physiological roles[1][2][3].
