Overview

Eukaryotic Deoxyribonucleic Acid (DNA) is the fundamental molecule of heredity, organized into complex linear chromosomes within the cell nucleus [NHGRI: DNA]. It serves as the master blueprint for all cellular processes, providing the templates for replication and the transcription of RNA required for protein synthesis. In the field of oncology, DNA is a critical therapeutic target for a wide range of cytotoxic chemotherapy agents [NCBI: Bookshelf NBK557404]. These drugs, including alkylating agents and intercalators, work by physically damaging the DNA structure or interfering with the enzymes that manage DNA topology, thereby preventing cancer cells from dividing. However, because these mechanisms often lack specificity for malignant cells, DNA-targeting therapies are frequently associated with significant systemic toxicities and the risk of inducing secondary mutations [PubMed: 25610363].

Mechanism of action

Drugs targeting eukaryotic DNA primarily act by forming covalent cross-links (alkylation), intercalating between base pairs to disrupt the double helix, or inducing single- and double-strand breaks, which collectively inhibit replication and transcription, leading to cell cycle arrest and apoptosis [StatPearls: Cancer Chemotherapy; PubMed: PMC113777].
