Evolutionarily conserved signaling intermediate in Toll pathway, mitochondrial (ECSIT) is a cytosolic adaptor protein involved in inflammatory signaling, embryonic development, and mitochondrial NADH:ubiquinone oxidoreductase (complex I) assembly and stabilization[1][2]. ECSIT is part of signaling pathways triggered by toll-like receptors, TGF-β, and BMP by forming complexes with other signaling proteins (e.g., TRAF6, TAK1/MAP3K7) that activate NF-κB and promote innate immunity and antiviral responses[1][3]. In mitochondria, ECSIT is essential for complex I assembly and thereby mitochondrial function; its perturbation results in defects in oxidative phosphorylation, cardiac pathology, and impaired immune responses[2][4]. ECSIT is also implicated in normal embryonic development and mesoderm formation[2]. There are currently no known drugs directly targeting ECSIT, but altered ECSIT expression is observed in some mitochondrial and cardiac diseases[3][4].
