"Excessive internal fire/toxicity" is a traditional Chinese medicine diagnostic concept, not a molecular entity. It refers to a pathological state characterized by "heat" or "fire" arising from the imbalance of yin and yang, usually involving excessive yang (heat, activity) relative to yin (cooling, moisture)[1][3][5]. Specific manifestations depend on the organ system affected (e.g., Heart fire, Liver fire, Stomach fire, Lung fire, Kidney fire). “Toxic-heat” (热毒, Rè Dú) represents a more severe, pathogenic form of fire, manifesting with symptoms such as swelling, redness, fever, pus, and pain, and is associated with internal or external pathogenic influences[3][7]. This framework is unique to TCM and guides both diagnosis and herbal intervention, but does not correspond to a discrete molecule, protein, enzyme, receptor, or other biomedical target[2][3][5]. ICD, IUPHAR, and other standard pharmacology databases do not recognize “internal fire/toxicity” as a therapeutic target. Rather, herbs and drugs in TCM categorized as clearing heat, draining fire, or detoxifying are used based on the syndrome’s clinical presentation, not to modulate a single molecular target[1][5]. The concept involves holistic body functions, not a physical receptor or protein. In summary: This is not a molecular or cellular “target” and should not be mapped as such for structured target-based research or drug development pipelines. It is a TCM pattern/syndrome, not a receptor, enzyme, transporter, or other druggable target class[1][3][5].
