Overview

Exported protein 1 (EXP1) is an essential membrane protein located in the parasitophorous vacuole membrane (PVM) of Plasmodium falciparum, the primary causative agent of malaria. It plays a critical role in maintaining the structural integrity of the PVM and is involved in the transport of nutrients and proteins between the host erythrocyte and the parasite. Beyond its structural role, EXP1 has been identified as a membrane-associated glutathione S-transferase (GS-transferase), which helps the parasite detoxify metabolic byproducts such as hematin. This enzymatic function makes it a significant target for antimalarial drugs, particularly artemisinin derivatives like artesunate, which bind to and inhibit EXP1. Inhibition of EXP1 leads to impaired detoxification and increased oxidative stress, ultimately resulting in parasite death. Because of its essentiality for the parasite's survival during the intra-erythrocytic stage, EXP1 is a focal point for understanding drug resistance mechanisms and developing next-generation antimalarials.
