Enzyme, Prenyltransferase, Protein modifying enzyme
01
Overview
Farnesyltransferase, CAAX box, alpha (FNTA) encodes the alpha subunit shared by protein farnesyltransferase and protein geranylgeranyltransferase type-1, which is essential for the prenylation of proteins containing a C-terminal CAAX motif. Preylation by these enzymes ensures the proper localization and function of various signaling proteins, including Ras and Rac1, and is implicated in cancer, metabolic, and neurodegenerative diseases. While FNTA is a validated therapeutic target, its related pseudogenes (including FNTAP1) are not known to encode active proteins nor are they clinically targeted[1][3].
Inhibition of protein prenylation (blocks attachment of farnesyl/geranylgeranyl to target proteins, e.g., Ras)
03
Biological functions
Protein prenylation (catalyzes transfer of farnesyl/geranylgeranyl group to CAAX-motif proteins)Membrane localization of target proteinsSignal transduction (downstream Ras, Rac1)Modulation of neuromuscular junction development (via RAC1 prenylation)Metabolic regulation (influences apolipoprotein A-I secretion)
Potential toxicity from interfering with essential signaling pathways (Ras, Rac1)Off-target effects (prenylation is necessary for many cellular functions; global inhibition may have broad impact)
06
Interacting drugs
Farnesyltransferase inhibitors such as tipifarnib, lonafarnib, and other experimental anticancer agents
07
Biomarkers
FNTA expression (as a marker for cancer progression/aggressiveness and chemoresistance)Prenylated Ras/Rac1 as indicators of pathway activation
