Fascin actin-bundling protein 1 (FSCN1) mRNA is the transcript encoding the Fascin-1 protein, which plays a pivotal role in organizing actin filaments into parallel bundles. These bundles are essential for the formation of filopodia and invadopodia, structures that drive cell motility and invasive behavior (UniProt P17677). While Fascin-1 is minimally expressed in normal epithelial tissues, its mRNA is significantly upregulated in various metastatic cancers, including breast, lung, and colorectal cancers, where it serves as a marker of poor prognosis (PubMed 30655540). Targeting the mRNA directly via RNA interference (RNAi) or antisense oligonucleotides (ASOs) allows for the knockdown of Fascin-1 protein production, effectively paralyzing the migratory machinery of cancer cells (PubMed 25661344). This approach has shown promise in preclinical studies for reducing tumor invasion and metastasis (PubMed 28813661). Current challenges in targeting FSCN1 mRNA include the development of efficient delivery systems to reach tumor cells and avoiding potential side effects in normal cells that express Fascin-1, such as mature dendritic cells (PubMed 22431510). By silencing the FSCN1 gene at the transcript level, researchers aim to provide a therapeutic strategy that complements existing treatments by specifically inhibiting the metastatic cascade.
