Overview

FERM, RhoGEF and pleckstrin domain-containing protein 1 (FARP1) is a multi-domain cytoplasmic protein containing a FERM domain, a Dbl homology (DH) domain, and two pleckstrin homology (PH) domains. It functions as a postsynaptic guanine nucleotide exchange factor, primarily activating the Rac1 GTPase, to regulate cytoskeletal dynamics and promote dendritic growth, synapse formation, and dendritic spine development in neurons. FARP1 interacts with cell surface proteins, including SynCAM 1 and PlexinA family receptors, via its FERM domain to mediate membrane localization and signal transduction. In addition to its critical role in neural development and synaptic organization, FARP1 also influences cell motility and invasion in non-neuronal cells by modulating Rho GTPase activity, with implications in cancer biology. Multiple transcript variants exist for this gene, and its dysfunction is associated with diseases such as cancer and congenital nystagmus.
