Overview

FERM, RhoGEF and pleckstrin domain-containing protein 2 (FARP2) is a multi-domain Dbl-family guanine nucleotide exchange factor (GEF) that activates Rac1 and, in some contexts, Cdc42, thereby mediating key processes in neuronal development and morphology, such as axon guidance, dendritic outgrowth, and actin cytoskeleton remodeling[1][2][3][4][7]. FARP2 is characterized by the presence of FERM, DH (Dbl-homology), and two PH (pleckstrin homology) domains, and its activity is regulated by auto-inhibition and potentially by interaction with plexin receptors and phosphorylation. The protein is implicated in pathways involving nervous system development, bone homeostasis (including osteoclast differentiation), and integrin-mediated cell adhesion. FARP2 has been linked to neurodevelopmental and skeletal disorders, but there are no known drugs or direct clinical biomarkers currently associated with it as a therapeutic target[1][2][4][7].
