Overview

Feruloyl-CoA 6'-hydroxylase 1 (F6'H1) is a plant enzyme belonging to the 2-oxoglutarate-dependent dioxygenase family that plays a critical role in the biosynthesis of coumarins, specifically scopoletin and its glucoside scopolin. In Arabidopsis thaliana, this enzyme catalyzes the essential ortho-hydroxylation of feruloyl-CoA, a key step in the phenylpropanoid pathway. This process is vital for the plant's response to nutrient stress, particularly during iron deficiency, as the secreted coumarins help mobilize and chelate iron from the soil for plant uptake. While F6'H1 is highly significant in agricultural biotechnology and plant physiology for improving crop nutrient efficiency, it is not currently recognized as a therapeutic target in human medicine. There are no known human drugs that interact with this plant-specific enzyme, and its primary interest lies in metabolic engineering and plant stress resilience.

Mechanism of action

Catalyzes the ortho-hydroxylation of feruloyl-CoA to 6'-hydroxyferuloyl-CoA using 2-oxoglutarate and molecular oxygen as co-substrates, which is the committed step in the biosynthesis of the coumarin scopoletin.
