Fidgetin-like protein 2 (FL2) mRNA is the messenger RNA transcript that encodes the FL2 protein, a microtubule-severing enzyme belonging to the AAA+ ATPase family [1, 5]. FL2 functions as a negative regulator of cell motility and axonal growth by selectively severing dynamic microtubules, thereby restricting the expansion of the microtubule cytoskeleton at the leading edge of migrating cells and the growth cones of neurons [1, 6]. In response to tissue injury, FL2 expression is typically upregulated, which can impede the natural regenerative process [2, 4]. Therapeutic targeting of FL2 mRNA using small interfering RNA (siRNA) or other gene-silencing technologies aims to transiently reduce FL2 protein levels to enhance tissue repair [5, 8]. Preclinical studies have demonstrated that silencing FL2 mRNA significantly accelerates cutaneous wound healing, promotes functional peripheral nerve regeneration, and improves recovery following spinal cord injury [1, 4, 9]. By decreasing the levels of this "brake" on the cytoskeleton, cells are better able to migrate and extend processes into damaged areas, facilitating faster closure of wounds and restoration of nerve function [5, 6].
