Overview

Filamenting temperature-sensitive mutant Z (FtsZ) is an essential cytoskeletal protein in Neisseria gonorrhoeae that serves as the primary coordinator of bacterial cell division. As a GTPase and structural homolog of eukaryotic tubulin, FtsZ polymerizes at the mid-cell to form the Z-ring, which acts as a scaffold for the recruitment of the divisome machinery required for septation. Because FtsZ is highly conserved across bacterial species and essential for viability, it has emerged as a prominent novel target for the development of antibiotics against multidrug-resistant (MDR) and extensively drug-resistant (XDR) gonorrhea. Experimental drugs like SMT-571 and various small-molecule inhibitors target FtsZ to disrupt Z-ring formation, leading to the arrest of cytokinesis and bacterial filamentation. While no FtsZ-targeting agents are currently in clinical use, the protein remains a high-priority candidate for both small-molecule drug discovery and AI-driven vaccine development.
