Frizzled-6 (FZD6) is a seven-transmembrane spanning protein that belongs to the Frizzled family of G protein-coupled receptors (GPCRs) [UniProt, P56747]. It serves as a receptor for Wnt proteins, playing a pivotal role in both canonical beta-catenin-dependent and non-canonical signaling pathways, such as the planar cell polarity (PCP) pathway [PubMed, PMID: 28213442]. FZD6 is essential for normal physiological processes including the development of hair follicles and nails, and mutations in the FZD6 gene are linked to autosomal recessive nail dysplasia [NCBI Gene, 8323; PubMed, PMID: 22305528]. In the context of oncology, FZD6 is frequently upregulated in various cancers, including breast, colorectal, and hematological malignancies, where it contributes to tumor progression, epithelial-mesenchymal transition (EMT), and chemoresistance [PubMed, PMID: 26876200]. Therapeutic targeting of FZD6 is an active area of research, primarily focusing on monoclonal antibodies and decoy receptors to disrupt Wnt-Frizzled interactions [PubMed, PMID: 25151356]. However, clinical development faces challenges due to the high homology among Frizzled family members and the potential for adverse effects on bone and intestinal homeostasis [PubMed, PMID: 25151356].
