Overview

Fructose-1,6-bisphosphatase (FBPase) is a critical regulatory enzyme in gluconeogenesis, catalyzing the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate. This reaction is irreversible and a key control point in glucose production. It is regulated by AMP and fructose 2,6-bisphosphate (inhibition) and ATP (activation). Two main isoforms exist: FBP1 (liver/kidney) and FBP2 (muscle). Deficiency causes hypoglycemia, while overactivity contributes to hyperglycemia in type II diabetes, making it a potential drug target. Increased expression may also play a role in appetite regulation.
