Fungal cell wall alpha-1,3-glucan and alpha-1,6-glucan are structural polysaccharides that contribute to the rigidity and protection of the fungal cell wall in various pathogenic species, such as Aspergillus fumigatus and Histoplasma capsulatum (PubMed: 17461667). Alpha-1,3-glucan, in particular, is often localized to the outermost layer of the cell wall, where it serves a critical role in virulence by masking beta-1,3-glucans from recognition by the host's Dectin-1 receptor, thereby preventing an effective immune response (PubMed: 18708524). While alpha-1,6-glucan linkages are less prevalent, they contribute to the overall complexity and branching of the glycan matrix in certain species. These molecules are considered attractive therapeutic targets because they are absent in humans, offering a high degree of selectivity for antifungal agents (PubMed: 11489122). Current research focuses on developing inhibitors of alpha-glucan synthases or utilizing enzymatic degradation via alpha-glucanases to compromise the fungal wall. Such interventions not only inhibit fungal growth but also unmask the fungus, allowing the host's innate immune system to identify and eliminate the pathogen more effectively.
