FYVE, RhoGEF and PH domain-containing protein 2 (FGD2) is a member of the FGD family of guanine nucleotide exchange factors (GEFs) that specifically activate the Rho family GTPase Cdc42 by facilitating the exchange of GDP for GTP[1][2][8]. FGD2 is highly expressed in antigen-presenting cells, such as B lymphocytes, macrophages, and dendritic cells, where it is involved in processes requiring actin cytoskeleton remodeling, including vesicle trafficking, immune cell migration, and antigen presentation[1][2][3]. The protein contains characteristic FYVE, RhoGEF, and pleckstrin homology domains, enabling binding to specific phosphoinositides and endosomal membranes as well as recruitment to actin-rich ruffles[2][6]. FGD2 is implicated in leukocyte signaling and immune regulation, and its dysregulation may be linked to cancer and immune-associated diseases[1][2][8]. Currently, there are no known clinically approved drugs directly targeting FGD2, nor are there established biomarkers or broadly recognized safety concerns unique to FGD2 as a drug target.
