Overview

G antigen (GAGE) is a member of the cancer-testis antigen (CTA) family, characterized by its restricted expression in normal tissues (primarily the testis and fetal ovary) and its aberrant expression in a wide variety of human cancers, including melanoma, lung, and ovarian carcinomas [1, 4, 21]. Due to its highly tumor-restricted expression pattern and immunogenicity, GAGE is a significant target for cancer immunotherapy, particularly in the development of peptide-based vaccines and adoptive T-cell therapies [1, 10, 19]. Biologically, GAGE proteins are thought to play roles in cell proliferation and the inhibition of apoptosis, potentially contributing to the aggressive phenotype and treatment resistance of certain tumors [11, 16, 23]. In clinical settings, the presence of GAGE is often used as a biomarker for molecular staging and prognosis, as its expression frequently correlates with poor survival and advanced disease stages [11, 28]. Therapeutic strategies targeting GAGE aim to harness the immune system to recognize and eliminate GAGE-positive malignant cells while sparing normal somatic tissues [1, 22].
