Overview

GPRC5D is an orphan G protein-coupled receptor in class C group 5, characterized as a seven-pass transmembrane receptor with a short extracellular N-terminal domain. It shows overexpression associated with plasma cell burden and genetic aberrations in multiple myeloma, with low expression in normal tissues, making it a selective surface target. Unlike some antigens, it is unlikely to be shed into serum, potentially reducing efficacy sinks for therapies. It is targeted by T-cell redirecting agents like bispecific antibodies and CAR T cells, achieving high response rates (≥64% overall, with many very good partial responses or better) in relapsed/refractory multiple myeloma patients.
