Overview

Pfs230 is a large, cysteine-rich surface protein of the malaria parasite Plasmodium falciparum, primarily expressed during the sexual stages of its life cycle [1, 11]. It belongs to the 6-cysteine protein family and plays a critical role in the fusion of male and female gametes and the interaction of male gametes with host erythrocytes within the mosquito midgut [1, 4, 12]. As a leading transmission-blocking vaccine (TBV) candidate, Pfs230 is designed to elicit antibodies in the human host that, when ingested by a mosquito, interrupt the parasite's development, thereby preventing further transmission to other humans [3, 11, 17]. Clinical development has focused on recombinant fragments, such as Domain 1 (Pfs230D1), which are often conjugated to carrier proteins like Pseudomonas aeruginosa Exoprotein A (EPA) to enhance immunogenicity [8, 16]. The primary mechanism of action involves complement-mediated lysis of gametes or steric hindrance of fertilization by vaccine-induced antibodies [11, 15, 20].
