Target intelligence / Profile preview
The Gamma-aminobutyric acid type A receptor (GABAA receptor) is a pentameric ligand-gated ion channel that serves as the primary mediator of fast inhibitory neurotransmission in the mammalian central nervous system (StatPearls, 2023). It is typically composed of five subunits—most commonly two alpha, two beta, and one gamma subunit—arranged to form a central chloride-selective pore (UniProt, 2024). Upon binding of the inhibitory neurotransmitter GABA, the receptor undergoes a conformational change that opens the pore, allowing chloride ions to flow into the neuron, which leads to hyperpolarization and a reduction in neuronal firing (NCBI, 2022). This receptor is a major pharmacological target for several classes of drugs, including benzodiazepines, barbiturates, and general anesthetics, which generally act as positive allosteric modulators to enhance GABAergic inhibition (PubChem, 2024). Clinically, GABAA receptor modulators are essential for treating conditions such as epilepsy, anxiety disorders, and insomnia, and are widely used in the induction and maintenance of anesthesia (PubMed, 2021). Dysregulation of GABAA receptor signaling is also implicated in the pathophysiology of neurodevelopmental disorders, depression, and schizophrenia (NIH, 2023). Furthermore, the receptor's diverse subunit composition allows for regional specificity in the brain, making it a versatile target for drug development (PubMed, 2022).
