Overview

The Gamma-aminobutyric acid type A receptor alpha-1 beta-2 delta subunit complex (GABA_A receptor α1β2δ) is a specific heteropentameric subtype of the ionotropic GABA receptor family [1, 2]. Unlike the more common synaptic αβγ receptors that mediate phasic (fast) inhibition, δ-containing receptors like α1β2δ are primarily located extrasynaptically and mediate tonic inhibition, providing a persistent inhibitory current that regulates the baseline excitability of neurons [11]. This receptor subtype is characterized by a high affinity for GABA but low efficacy in the absence of modulators, often being described as effectively silent until recruited by endogenous or exogenous agents [6, 11]. It is a primary target for neurosteroids such as allopregnanolone and synthetic analogs like brexanolone and zuranolone, which act as positive allosteric modulators to enhance inhibitory tone [9, 11]. Dysregulation of these receptors is implicated in various neuropsychiatric conditions, including postpartum depression, major depressive disorder, and epilepsy [2, 12]. Consequently, the α1β2δ receptor is a critical therapeutic target for restoring GABAergic balance in the central nervous system [10, 11].
