The Gamma-aminobutyric acid type A receptor subunit gamma-1 (GABRG1) is a protein that forms part of the heteropentameric GABA-A receptor, the primary inhibitory neurotransmitter receptor in the mammalian central nervous system (Wikipedia, 2024; UniProt, 2024). Unlike the ubiquitous gamma-2 subunit, GABRG1 expression is more localized, with high levels in regions such as the amygdala, hypothalamus, and substantia nigra, suggesting specialized roles in emotional regulation and endocrine function (NIH, 2009; Creative BioLabs, 2024). Receptors containing the gamma-1 subunit possess a unique pharmacological profile; they generally show lower affinity for classical benzodiazepines and are notably resistant to the antagonist effects of flumazenil (ResearchGate, 2016). Genetic studies have strongly linked GABRG1 variants to alcohol dependence, anxiety disorders, and eating disorders, highlighting its potential as a precision therapeutic target (NIH, 2008; Maayanlab, 2024). Developing selective modulators for gamma-1-containing receptors could offer new avenues for treating neuropsychiatric conditions with a reduced risk of the side effects associated with non-selective GABAergic drugs (CureGABA, 2021).
