Overview

Gastrin mRNA is the messenger RNA transcript of the GAST gene, which encodes the preprogastrin precursor protein (UniProt: P01350). This precursor is processed into various bioactive forms of gastrin, primarily gastrin-17 and gastrin-34, which are essential for stimulating gastric acid secretion from parietal cells and promoting the growth of the gastric and colonic mucosa (NCBI Gene ID: 2520). In pathological states, such as gastrinomas or certain gastrointestinal cancers, GAST mRNA is often overexpressed, leading to hypergastrinemia and accelerated tumor cell proliferation (PMID: 15188258). Therapeutic strategies targeting GAST mRNA, such as antisense oligonucleotides or small interfering RNAs, aim to silence the expression of gastrin at the pre-translational level (PMID: 10446449). This approach is intended to control acid hypersecretion in Zollinger-Ellison syndrome and inhibit the trophic effects of gastrin on hormone-dependent malignancies (PMID: 11506514). While no mRNA-targeted therapies for gastrin are currently FDA-approved, research continues into their potential for precision oncology and managing refractory hypersecretory disorders.
