Overview

GC-rich DNA binding sites, commonly known as GC boxes, are critical cis-regulatory elements found in the promoter regions of numerous housekeeping and tissue-specific genes (Suske, G., 1999, Gene). These sequences, typically characterized by the consensus motif 5'-GGGGCGGGG-3', serve as the primary docking sites for the Specificity Protein (Sp) and Kruppel-like factor (KLF) families of transcription factors (Safe, S., & Abdelrahim, M., 2005, Eur J Cancer). By facilitating the recruitment of the transcriptional machinery, these sites play a fundamental role in regulating essential cellular processes such as proliferation, differentiation, and apoptosis. In many cancers, the over-activation of genes regulated by these GC-rich sites contributes to tumor progression, angiogenesis, and resistance to therapy (Vizcaíno, C., et al., 2015, Gene). Consequently, these DNA sequences have emerged as therapeutic targets for minor groove-binding agents like Mithramycin A, which competitively inhibit transcription factor binding (Previdi, S., et al., 2010, PLoS One). Targeting these sites offers a strategy to downregulate multiple oncogenic pathways simultaneously, although clinical utility is often limited by systemic toxicity and a narrow therapeutic window (PubChem, CID 16218737).
