Overview

Site-specific integration into the genome refers to the precise insertion of genetic material at defined locations within a host organism’s DNA. This process is mediated by specialized enzymes, most notably site-specific recombinases or integrases, which recognize and act upon short, specific DNA sequences known as recombination sites. Unlike random integration methods—which can lead to unpredictable gene expression and potential disruption of essential genes—site-specific approaches enable controlled and predictable genetic modifications. Commonly used systems include Cre-loxP, PhiC31 integrase, and FLP-FRT.
