Overview

Genome-derived Neisseria antigen 2091 (GNA2091) is a highly conserved lipoprotein found in Neisseria meningitidis and is a key component of the multicomponent serogroup B meningococcal vaccine, 4CMenB (Bexsero) [1, 3]. It is localized to the periplasmic side of the bacterial outer membrane and functions as an analogue of the Escherichia coli YraP protein, playing a critical role in outer membrane biogenesis and the assembly of outer membrane proteins (OMPs) [1, 4]. Research indicates that GNA2091 is essential for maintaining membrane stability and zinc homeostasis, and its absence leads to increased outer membrane vesicle production and attenuated virulence [2, 4]. In the 4CMenB vaccine, GNA2091 is expressed as a fusion protein with Factor H-binding protein (fHbp) to improve the stability and overall immunogenicity of the vaccine [6, 8]. While antibodies specifically targeting GNA2091 may not be directly bactericidal due to its periplasmic location, the protein's inclusion is vital for the vaccine's broad protective efficacy against various meningococcal strains [1, 6]. Additionally, GNA2091 homologs in Neisseria gonorrhoeae suggest a potential role in cross-protection against gonorrhea [8, 9].
