Germin-like protein (Citrus sinensis) (Cit s 1)

Target
Cit s 1
Molecular classification
Germin-like protein, Plant allergen, Cupin superfamily
01

Overview

Germin-like protein Cit s 1 is a glycoprotein allergen isolated from sweet orange (Citrus sinensis). It belongs to the germin-like subfamily within the cupin superfamily of plant proteins. Cit s 1 is identified as a 24 kDa glycoprotein that binds specific IgE in more than 60% of orange-allergic individuals, although only a small subset have positive clinical responses (such as skin reactions) when exposed to purified Cit s 1. Its IgE-binding is heavily dependent on complex N-glycan structures, which can result in false positive diagnosis of orange allergy if based solely on in vitro IgE testing. Cit s 1 does not currently serve as a therapeutic target; its primary significance is as a component allergen in food allergy diagnosis and research.

Biological functions

Putative role in plant defense (typical germin-like protein function)acts as an allergen in humans
03

Disease associations

Allergic reaction (food allergy, specifically orange allergy)
04

Safety considerations

False positives in in vitro IgE testing due to strong IgE binding to glycans (not always clinically relevant)rarely elicits strong clinical reactions despite IgE binding
05

Biomarkers

Serum specific IgE to Cit s 1 (used experimentally/clinically to detect orange allergy)
