GHRLOS is a long non-coding RNA (lncRNA) gene that is transcribed antisense to the ghrelin gene (GHRL) on chromosome 3p25.3[1][2]. It spans approximately 44 kb and yields multiple, extensively spliced RNA isoforms that are capped, polyadenylated, and tissue-specifically expressed, especially in the brain, thymus, testis, ovary, and uterus[1][2]. GHRLOS overlaps both the GHRL gene and a variant of the SEC13 gene, which may allow it to regulate transcription or translation of these genes through antisense mechanisms[1]. GHRLOS has no protein-coding potential and is highly divergent between species, suggesting rapid evolutionary change[1]. Functionally, GHRLOS is thought to play a regulatory role in the ghrelin axis and possibly other gene networks. Its expression is altered in colorectal cancer, where reduced GHRLOS levels are associated with metastasis and poor prognosis, indicating its potential utility as a disease biomarker[3].
