The GLI family zinc finger 3 (GLI3) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the GLI3 transcript, which encodes a key transcription factor in the Hedgehog signaling pathway (NCBI Gene: 2737). This region serves as a docking site for various microRNAs (miRNAs) and RNA-binding proteins that modulate the stability and translation efficiency of the GLI3 mRNA. In many cancers, such as lung cancer and medulloblastoma, dysregulation of the Hedgehog pathway leads to aberrant GLI3 expression, often facilitated by the loss of inhibitory miRNA binding to the 3'-UTR (PubMed: 28656245). Consequently, the GLI3 mRNA 3'-UTR is considered a viable therapeutic target for RNA-based interventions, including miRNA mimics or antisense oligonucleotides designed to restore normal regulatory control. By specifically targeting this region, researchers aim to downregulate GLI3 protein levels, thereby inhibiting tumor growth and progression (PubMed: 30210156). Beyond oncology, mutations or regulatory defects involving GLI3 are linked to developmental disorders like Greig cephalopolysyndactyly syndrome (PubMed: 17033630).
