Overview

Glucosyltransferase C (GtfC) is a key extracellular enzyme produced by the bacterium Streptococcus mutans, which is widely recognized as the primary initiator of dental caries (UniProt: P11047). This enzyme is responsible for the synthesis of both water-soluble and water-insoluble glucans from dietary sucrose (PubMed: 11133465). These glucans serve as a sticky scaffold that facilitates the firm attachment of S. mutans and other oral bacteria to the tooth surface, leading to the formation of a robust dental biofilm or plaque. The insoluble glucans produced by GtfC are particularly critical for the structural integrity of the biofilm, creating a protected environment where organic acids produced by bacterial fermentation can demineralize tooth enamel (PubMed: 21844338). Because GtfC is essential for the transition of S. mutans from a planktonic state to a pathogenic biofilm, it is a major target for pharmacological intervention. Inhibiting GtfC activity can effectively reduce the cariogenic potential of the oral microbiome without the use of broad-spectrum antibiotics, thereby preserving the natural balance of oral flora while preventing tooth decay (PubMed: 25601155).
