The NMDA receptor (N-methyl-D-aspartate receptor) is a subtype of ionotropic glutamate receptors that plays a central role in excitatory neurotransmission, synaptic plasticity, and neurodevelopment in the mammalian central nervous system. It is an obligate heterotetramer typically composed of two glycine-binding GluN1 subunits and two glutamate-binding GluN2 subunits. Activation requires co-agonist binding of glycine and glutamate, and channel opening is voltage-dependent due to Mg²⁺ block. The receptor mediates Ca²⁺ influx critical for various physiological processes, but dysregulation can lead to excitotoxicity and involvement in various neurological disorders.
