Overview

Glycine-extended gastrin-17 (G17-Gly) is a non-amidated, 18-amino acid precursor of the mature hormone gastrin-17, functioning as a significant biological entity with distinct trophic effects [4, 5]. While mature gastrin regulates acid secretion via the CCK2 receptor, G17-Gly acts as a potent mitogen and growth factor in the gastrointestinal tract, promoting cell proliferation and inhibiting apoptosis through the activation of signaling pathways like JAK2/STAT3, PI3K/Akt, and JNK [2, 8]. Elevated concentrations of G17-Gly are frequently detected in patients with colorectal, pancreatic, and gastric cancers, where it facilitates tumor progression via autocrine and paracrine mechanisms [1, 7]. Notably, G17-Gly exerts its proliferative effects through a putative high-affinity receptor that is distinct from the classical CCK2 receptor, making it a selective target for oncological intervention [3, 7]. Therapeutic strategies such as G17DT (Gastrimmune) utilize an immunogenic approach to induce neutralizing antibodies that target G17-Gly, thereby blocking its ability to stimulate hormone-dependent tumor cell expansion [5, 9]. This peptide also plays a synergistic role with amidated gastrin in regulating gastric acid secretion and maintaining the integrity of the gastric epithelium [10, 11].
