Overview

The glycine site is a modulatory binding site located on the GluN1 subunit of the NMDA receptor, a subtype of ionotropic glutamate receptors. It is strychnine-insensitive and distinct from classical inhibitory glycine receptors. This site requires either glycine or D-serine as a co-agonist, along with glutamate, for NMDA receptor activation. The glycine/D-serine binding acts allosterically to facilitate NMDA receptor function, which is critical for synaptic plasticity, learning, memory formation, and neurodevelopment. Its dysfunction is implicated in neurological disorders.
