Overview

The Glycogen synthase kinase 3 beta (GSK3B) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the GSK3B transcript that governs the expression levels of the GSK3B protein (UniProt P49841). This region contains numerous binding sites for microRNAs (miRNAs) such as miR-26a, miR-101, and miR-129, which collectively modulate mRNA stability and translation efficiency (PubMed: 24603440, 25119378). GSK3B itself is a proline-directed serine/threonine kinase involved in diverse cellular processes, including glycogen metabolism, Wnt signaling, and cellular proliferation. Dysregulation of GSK3B expression, often mediated by alterations in its 3'-UTR, is strongly associated with the pathogenesis of Alzheimer's disease, mood disorders, and various cancers (PubMed: 26823744). Therapeutic strategies targeting the GSK3B mRNA 3'-UTR, such as antisense oligonucleotides or miRNA mimics, aim to fine-tune GSK3B protein levels rather than completely inhibiting its enzymatic activity. This approach offers a potential advantage in treating chronic conditions where precise modulation of signaling pathways is required to avoid systemic toxicity associated with global kinase inhibition.
