Glycosphingolipids (GSLs) are a diverse class of membrane lipids composed of a hydrophobic ceramide tail and a hydrophilic glycan head group [1]. They are primarily located on the outer leaflet of the plasma membrane, where they organize into microdomains known as lipid rafts to facilitate signal transduction and cell-cell communication [2]. GSLs are critical for various physiological processes, including neural development and immune response modulation [1]. Pathologically, the accumulation of GSLs due to genetic deficiencies in lysosomal hydrolases leads to lysosomal storage diseases such as Gaucher and Fabry disease [3]. In oncology, certain GSLs like the ganglioside GD2 are overexpressed on the surface of neuroblastoma and melanoma cells, serving as specific targets for immunotherapy [4]. Therapeutic interventions include substrate reduction therapies (SRTs) like eliglustat, which inhibit GSL synthesis, and monoclonal antibodies like dinutuximab, which target tumor-associated GSLs [3,4].
